人血白蛋白是一种在血液存在的蛋白质,可用于一系列疾病的治疗,其中就包括肝衰竭和创伤性休克。过去也有尝试在猪体内产生人血白蛋白,但是由于无法避免猪源的白蛋白的产生,这使得分离与提纯人源血白蛋白具有很大挑战。

中国科学家张普民和他的研究团队,使用CRISPR/Cas9基因编辑系统,在猪受精卵基因组中用于产生猪白蛋白的区域,插入了一段可以合成人类白蛋白的DNA片段。在猪基因组中嵌入的等位基因位于猪白蛋白合成基因的下游。研究者认为,这可以让猪产生人源的血清白蛋白的同时,却不产生猪源白蛋白,这样就可以避免人源血清白蛋白和猪源白蛋白的混合。

研究者植入了300个基因组编辑后的受精卵到10只母猪体内,这些受精卵都携带有插入人血白蛋白的等位基因。5只猪成功怀孕,产下了16只巴马猪仔。研究者发现这16只猪仔来自携带人源白蛋白基因的受精卵,并且它们的血液中能检测到人血白蛋白。不过,猪仔体内的人血白蛋白含量有所不同。进一步的繁殖实验表明,通过CRISPR/Cas9基因修饰工具插入的人源基因可以传给这些猪仔的后代。
