在一项新研究中,澳大利亚阿德莱德大学的研究人员揭示了在中国临床上使用多年的中药——复方苦参注射液(CKI)如何杀死癌细胞,相关研究结果于近日发表在《Oncotarget》杂志上,是首个在不破坏结构的基础上分析中医分子作用机制的研究。

CKI已在中国临床上使用了15年,通常被用于治疗多种实体肿瘤,也被当做西医化疗的辅助治疗,但它如何起作用至今仍未清楚。然而,由于传统中药是植物次生代谢产物的复杂混合物,利用系统方式来探索其分子机制十分有必要。

本研究的引领者为David Adelson教授,他说,“大多数的中药是基于中国上千年的使用经验,通常有大量的证据表明这些药物的治疗益处,但人们并未了解它们的作用机制。如果打破并测试许多传统中药的成分,我们会发现个别化合物本身并没有太多的活性物质。中药是多种有效化合物的组合,这意味着副作用也比较少。”

利用高通量测序技术来揭秘中医如何杀死癌细胞

在该研究中,研究人员利用MCF-7人类乳腺癌细胞系作为体外研究模型,用于识别CKI诱导的基因表达变化。研究人员将两种不同浓度的CKI(1 and 2 mg/mL总生物碱类)作用于乳腺癌细胞,作用时间为24小时和48小时。

研究人员测定了CKI对细胞增殖和细胞凋亡的影响,并利用Illumina的RNA-seq技术来测定CKI处理24小时和48小时后的细胞转录组。

结果发现,CKI抑制了乳腺癌细胞的增殖,并以剂量依赖性的方式诱导细胞凋亡。研究人员应用了一系列的转录组分析方法来确定CKI的候选抗癌分子机制。多途径干扰和细胞周期识别是CKI在乳腺癌细胞中的潜在靶向途径。CKI还可能通过p53的独立机制来诱导乳腺癌细胞凋亡。此外研究人员还确定了新的 lncRNAs,并表明在CKI的作用下,很多lncRNAs进行了表达。

Adelson教授说,“由CKI触发的基因表达模式与西医化疗影响的生物学通路一致,但在同种通路中所作用的基因不同。这些基因调节细胞分裂和死亡周期,这似乎意味着CKI改变了细胞周期的调控方式,将癌细胞推向死亡道路,从而杀死它们。”

Adelson教授补充说,“这项技术可以用来分析其他重要的分子机制,为中药应用至西医打开大门。”
