J Ethnopharmacol:逍遥丸基于肠道微生物群的代谢产物通过抑制大脑中的脂肪酸酰胺水解酶水平来改善抑郁症

从体内外两个层面探讨逍遥丸(XYPs)潜在的抗抑郁机制。

背景:中药通常以口服剂型制备,使中药在口服后与肠道微生物群相互作用,这可能会影响中药的治疗效果。逍遥丸(XYPs)是中国治疗抑郁症的常用中药。然而,由于其复杂的化学成分,其生物学基础仍处于初级阶段。该研究旨在从体内外两个方面探讨XYPs潜在的抗抑郁机制

方法:XYPs由8种中药组成,包括柴胡、当归、芍药、茯苓、甘草、薄荷、白术、生姜,比例为5:5:5:4:1:5:5:5。建立慢性不可预测轻度应激(CUMS)大鼠模型。之后,进行蔗糖偏好试验(SPT)以评估大鼠是否抑郁。治疗28天后,进行强迫游泳试验和SPT,以评估XYPs的抗抑郁疗效。取出粪便、大脑和血浆进行16SrRNA基因测序分析、非靶向代谢组学肠道微生物群转化分析。

结果:该结果揭示了XYPs影响的多种途径。其中,通过XYPs处理,大脑中脂肪酸酰胺的水解降低最为显著。此外,在CUMS大鼠的血浆和大脑中发现了主要来源于肠道微生物群的XYPs代谢产物(苯甲酸、甘草素、甘草次酸和柴胡皂苷元D),并可抑制大脑中脂肪酸酰胺水解酶(FAAH)的水平,这有助于XYPs的抗抑郁作用。

结论:非靶向代谢组学结合肠道微生物群转化分析揭示了XYPs潜在的抗抑郁机制,进一步支持了肠脑轴理论,为药物发现提供了有价值的证据。
