“天然灭蚊剂”BinAB结构发布

英国《自然》杂志29日在线公开的一篇传染病学研究报告,发布了细菌毒素BinAB的结构。BinAB是一种对控蚊非常重要的天然杀虫剂,其结构阐明了这一毒素此前未明的生物学功能。该研究还表明,这一方法也可用于对其他受晶体小尺寸限制的杀虫蛋白进行结构解析。

蚊子可传播疟疾、登革热及近期流行的寨卡病毒,是严重危害人类健康的昆虫之一。蚊子的幼虫孑孓由蚊卵在水中孵化而成,属蚊子由卵成长至蛹的中间阶段。人类迄今已使用了非常多的控蚊方案,包括自然产生的蛋白质晶体形式的灭孑孓剂,如细菌毒素BinAB等。

BinAB是由球形赖氨酸芽孢杆菌产生的,对孑孓有毒,但对人类和其它动物无害,已被一些国家用于控制蚊子数量。但由于BinAB的晶体非常小,解析其结构十分困难,截至目前,人们对BinAB毒素及其作用机制的理解仍十分有限。

法国结构生物学研究所科学家雅克-菲利普·卡勒帝亚及其同事,此次使用了一种名叫串行飞秒激光晶体学的技术。该技术利用了一种强大的激光源——X射线自由电子激光 (XFEL),以前所未有的清晰度解析了细菌毒素BinAB结构,最终发现了可能与其灭孑孓功能有关的酸碱度敏感开关和碳水化合物结合模块,以及维持细菌毒素BinAB效力的分子间相互作用。

这一研究成果将有助于增强BinAB灭孑孓剂的效力,扩大其使用范围。更重要的是,研究同时表明,新方法也能解析其他受晶体小尺寸限制的抗疟疾和杀虫蛋白质结构。
