我们提取了MDD、2型糖尿病、CAD和心力衰竭的摘要级数据，这些数据来自于欧洲后裔已发表的大型全基因组相关性研究。在全基因组显著性水平上，MDD有96个SNPs, 2型糖尿病有202个SNPs，冠心病有44个SNPs，心力衰竭有12个SNPs被认为是工具变量(p < 5×10−8)。主要分析采用随机效应反方差加权法。

在bonferroni校正的显著水平上，MDD的遗传倾向与2型糖尿病和CAD显著相关。2型糖尿病患者的ORs分别为1.26 (95% CI 1.10, 1.43);p = 6×10−4和1.16 (95% CI 1.05, 1.29;(p = 0.0047) MDD的loge发生率每增加1个单位。MDD与心力衰竭之间存在暗示相关性(或1.11[95%可信区间1.01,1.21];p = 0.033)。在反向磁共振分析中，我们发现支持心脏代谢疾病对MDD风险的因果影响的证据有限。

本研究进一步证实了MDD是2型糖尿病和冠心病的潜在危险因素。MDD是否与心力衰竭有因果关系尚需进一步研究。
