基因表达的表观遗传调控发生在被称为拓扑关联域(TADs)的离散空间染色体单元内,但大多数基因的精确的空间要求尚未明确;该点对与癌症相关的基因特别有意义。

Daniel Schuetzmann等人采用高分辨率的染色质构象捕获测序,尝试在健康单核细胞和急性髓系白血病(AML)细胞中绘制编码关键髓系转录因子PU.1的人基因座的三维(3D)结构图。

研究人员确定了一个动态的大约75kb大小的单位(SubTAD)作为基因组区域,该区域内PU.1基因调控元件之间的空间互作发生在髓系分化过程中,并在AML中被破坏。在该SubTAD内,空间染色质互作的合理启动需要PU.1自动调控以及招募染色质衔接蛋白LDB1(LIM结构域结合蛋白1)。但是,这些空间互作一旦发生,LDB1就可不依赖PU.1自动调节而稳定它们。

因此,研究人员得出结论,PU.1通过启动稳定的染色质环,以“打了就跑”的方式自动调节其表达,从而形成转录活跃的染色质结构。
