索元生物抗肿瘤创新药的突破性研究成果将在美国血液学年会展示

精准医疗领军企业索元生物今日宣布:将在2018年12月1日至12月4日举行的美国血液学年会(ASH)上将以学术壁报形式展示两项关于DB102(enzastaurin)的新进展。第一张学术壁报通过对既往弥漫性大B细胞淋巴瘤(DBLCL)维持疗法的一个三期临床试验的回顾性分析发现一个全新的可预测DB102药物疗效的生物标记物Denovo Genetic Marker 1 (DGM1), 并且此DGM1

第一张学术壁报通过对既往弥漫性大B细胞淋巴瘤(DBLCL)维持疗法的一个三期临床试验的回顾性分析发现一个全新的可预测DB102药物疗效的生物标记物Denovo Genetic Marker 1 (DGM1), 并且此DGM1与DB102药效的相关性在另外一个独立DLBCL一线治疗的二期临床试验中得到验证。在两项研究中,与DGM1阴性患者相比,DGM1阳性患者的总生存期(OS)显著延长。根据这些结果,索元生物启动了在DLBCL初治患者中在生物标记物指导下的关键性的临床三期试验(ENGINE试验)。


第二张学术壁报为北京大学肿瘤医院朱军教授实验室在临床前研究中发现DB102与BTK抑制剂ibrutinib联合使用可产生协同性的抗肿瘤活性。 相比于使用单药,在ABC及GCB DLBCL细胞系中联合添加DB102和ibrutinib时,可对肿瘤细胞的存活及扩增产生持久的协同性抑制作用。两药的联合治疗还展现了协同性的抑制肿瘤细胞的转移、侵蚀、以及相关联的主要信号通路。这些结果为未来的DB102与BTK抑制剂如ibrutinib的临床试验奠定了基础。

关于DB102

DB102 (Enzastaurin)是一类全球首创口服小分子丝氨酸/苏氨酸激酶抑制剂,作用于PKCβ、PI3K和AKT等肿瘤领域的关键靶点上。已有3000多例血液肿瘤和实体瘤受试者接受该药的治疗。DB102最初由礼来公司开发,索元公司整体收购了DB102项目,目前拥有该药全球开发、生产和销售的全部权利。DB102在弥漫性大B细胞淋巴瘤和脑胶质瘤两个领域已获得美国和欧洲的孤儿药认证。

关于ENGINE临床试验

ENGINE试验是一项全球、随机、双盲、经批准的临床试验,评价Enzastaurin联合R-CHOP对比R-CHOP在携带全新基因生物标记物DGM1的初治高危弥漫性大B细胞淋巴瘤受试者中的有效性和安全性。本试验将入组235例受试者,主要终点为评估携带DGM1的受试者的总生存期。
