Oncol Rep:过氧化氢可通过诱导细胞死亡和G1期阻滞来抑制肺癌细胞的生长

过氧化氢(H2O2)经常用于培养细胞以诱导氧化应激。本研究调查了外源性H2O2对Calu-6和A549肺癌细胞的作用。MTT测定显示,H2O2抑制Calu-6和A549细胞的生长,24小时IC50值分别为~50和100μM。用H2O2处理的细胞表现出细胞在G1期停滞。H2O2呈剂量依赖性地增加了这些细胞的死亡(台盼蓝阳性)和膜联蛋白V-FITC染色细胞的数量,伴随着Bcl-2和前半胱天冬酶-3水平

所有测试的半胱天冬酶抑制剂H2O2处理的Calu-6和A549细胞中(泛半胱天冬酶的Z-VAD,caspase-3的Z-DEVD,caspase-8的Z-IETD和caspase-9的Z-LEHD)降低了亚G1和膜联蛋白V-FITC染色细胞的百分比。然而,半胱天冬酶抑制剂不能明显阻止H2O2处理的肺癌细胞中MMP(ΔΨm)的丧失。

总之,H2O2通过细胞死亡和G1期阻滞抑制Calu-6和A549肺癌细胞的生长。H2O2诱导的细胞死亡是由细胞死亡以及半胱天冬酶依赖性细胞凋亡引起的。
