线粒体是产生大部分细胞ATP的动态细胞器,其也会涉及许多其他的细胞功能,例如Ca 2+离子的信号传导,细胞分化,凋亡,细胞周期,以及细胞生长。

线粒体动力学的一个关键过程是线粒体融合,其由mitofusinsMFN1MFN2)和OPA1催化。线粒体外膜蛋白MFN2在维持线粒体代谢,胰岛素信号传导,和导致神经退行性疾病的突变中起着相关作用。因此,对参与线粒体动力学的蛋白质的调节已经成为潜在的药物靶点。

近日,来自巴塞罗那大学的研究人员发文报导,其通过高通量筛选,识别了能够促进线粒体以MFN1 / MFN2依赖性方式延伸的小分子。

对其作用模式的详细分析揭示了嘧啶代谢与线粒体动力学之间的联系,这是以前没有被报导过的。研究人员的数据表明,嘧啶生物合成与线粒体动力学之间存在着联系,其在以嘧啶合成损失为特征的应激条件下维持细胞存活。
