日本理化学研究所(RIKEN,RIkagaku KENkyusho/Institute of Physical and Chemical Research)领导的FANTOM项目团队(fantom.gsc.riken.jp)在十年前发现并开创了非编码RNA研究领域,首次揭示了哺乳动物基因组转录过程的复杂性。 

十年之中,FANTOM项目团队继续着对非编码RNA起源和功能的研究。

FANTOM对于非编码RNA的最新研究进展发表在最新一期Nature杂志上,标题为“An atlas of human long non-coding RNAs with accurate 5′ ends”。

在这一最新的研究中,FANTOM团队建立了一个全面的人类非编码长链RNA图谱,基本对既有的基因表达模型进行了深度的改进,让其他研究人员能够更好地对RNA的多样性与不同功能进行评估。

目前大多数与RNA转录图谱绘制相关的研究相对过分依赖准确度较低的RNA转录产物起始或5'端测序。为了克服这个限制,FANTOM团队使用了一种称为基因表达Cap分析(CAGE)的新技术。该技术用于构建具有更为精确的5'末端人非编码长链RNA图谱,可以精确定位相关RNA在基因组中的起始位置。

FANTOM的最新图谱包含了27,919个非编码长链RNA,首次阐明并总结了它们在主要人类细胞类型和组织上的表达模式。通过该图谱与基因组遗传数据的综合分析,研究人员发现人类基因组中存在的19,175个功能相关非编码长链RNA,这意味着人类基因组内可能存在大约20,000个与蛋白编码基因同样会对疾病发生发展造成影响的非编码长链RNA。

“学界对于我们基因组中数以千计非编码长链RNA存在着激烈的争论。这些RNA究竟是像蛋白编码DNA一样影响着我们的性状,还是仅仅作为转录翻译的副产物存在于细胞中,一直都是一个令人费解的问题。”文章的作者,RIKEN生命科学技术中心(Center for Life Science Technologies ,CLST)高级访问学者,西澳大利亚大学哈里珀金斯医学研究所的Alistair Forrest教授表示。 “通过整合和改进基因的表达模型,结合进化和遗传研究的相关数据,我们发现这些非编码长链RNA中的大多数看起来似乎是拥有某些功能的。同时,我们发现其中将近2,000个非编码长链RNA与疾病和其他相关遗传性状有着潜在的联系。”

“有趣的是,”文章的第一作者,CLST的Chung-Chau Hon博士表示,“我们发现大多数非编码长链RNA似乎是从增强子中转录产生的,这一发现加深了我们对非编码长链RNA主要异质起源的理解。”
