癌症研究着Raffaella Sordella博士和他的同事在美国国家科学院院刊的一篇报道提出了p53的“堂兄妹”即p53-psi 的新发现,这是一个不为人知的p53蛋白的变异种,它遗传了相同的基因,在人体中被称为TP53 ,它是一种以其他形式表现的p53。

Sordella和他的同事观察当p53-psi表达时,它降低了黏钙蛋白的表达量,黏钙蛋白通常的作用是使细胞与上皮组织保持接触,肺里层和其他许多身体器官都是由上皮组织构成的。这是伴随着关键细胞标志物与肿瘤的侵袭性和转移潜能有关联的表达(这些是EMT的标记,EMT即上皮细胞向间充质细胞转化)。Sordella和他的同事发现在肺癌早期预后不良的情况下p53-psi的水平有所升高。

经调查发现,在细胞膜的能量工厂线粒体中通过刺激新一代氧化分子-活性氧(ROS)后,p53-psi与cyclophillin D蛋白产生的相互作用引起了较强的增长效果。

p53-psi被发现在肿瘤和受伤的组织中固有表达。Sordella说,有可能更熟悉的p53突变体与肿瘤生长和转移相关联,细胞程序通过无数次的进化愈合可能被p53突变体“劫持”使癌症扩散失控。

该研究小组目前正在调查研究在伤口愈合中p53-psi的作用机理。研究者希望通过后期的深入研究揭示p53突变的机制来帮助理解癌症发生的过程。
