α-突触核蛋白（α-syn）的病理积累和聚积是帕金森病（PD）的基础。 但是至今为止，病理性α-syn导致PD神经变性的分子机制尚不清楚。
最近，研究人员发现病理性α-syn激活聚（腺苷5'-二磷酸核糖）（PAR）聚合酶-1（PARP-1），而PAR的产生加速病理性α-syn的形成，通过parthanatos导致细胞死亡。
PARP抑制剂或PARP-1的遗传缺失阻止了病理性α-syn的毒性。在前馈环路中，PAR将病理性α-syn转化为毒性更大的蛋白。PD患者的脑脊液和脑中PAR水平升高，提示PARP活化在PD发病机制中起作用。
因此，旨在抑制PARP-1活化的策略可以作为一种疾病调节疗法，以防止PD中多巴胺神经元的丧失。
