Blood:ECM-整合素受体轴在原发性骨髓纤维化巨核细胞异常增殖中的作用

整合素a5b1的a5亚基在PMF的JAK2V617F+巨核细胞中表达上调,导致对纤连蛋白的粘附性增强。 无论是在体内还是体外,抗体介导的a5亚基抑制均可阻止JAK2V617F+巨核细胞谱系扩增。

细胞外基质(ECM)过度积累是在原发性骨髓纤维化(PMF)骨髓(BM)环境中的一个标志。由于细胞具有通过整合素受体粘附到周围ECM的能力,Matsuura等人验证了ECM-整合素受体轴异常导致JAK2V617F+PMF骨髓巨核细胞增多的假设。PMF患者骨髓基质细胞分泌的ECM蛋白纤维连接蛋白(FN)与纤维化和疾病严重程度相关。

在本研究中,研究人员发现Vav1-hJAK2V617F转基因小鼠(JAK2V617F+)具有与巨核细胞增多症和纤维化相关的高BM FN含量表现。此外,与野生型对照相比,JAK2V617F+小鼠的巨核细胞表面的a5b1整合素的a5亚单位(巨核细胞上的主要的FN受体)的表达量增加,增强了与FN的粘附。

无论是在体内还是体外,用a5亚单位的抑制性抗体均可阻断其与FN的粘附,进而有效地降低了CD41+JAK2V617F+巨核细胞的比例。与在小鼠上的发现相一致,来自患者的JAK2V617F+巨核细胞的a5亚基的表达水平也升高了,而且a5亚基中和抗体同样减少了对来自CD34+细胞的FN和巨核细胞的粘附。

综上所述,本研究揭示了FN-a5b1整合素在JAK2V617F+PMF中对巨核细胞增殖的前所未闻的贡献。
